{"id":116,"date":"2021-11-05T13:02:27","date_gmt":"2021-11-05T12:02:27","guid":{"rendered":"https:\/\/docenciamicrobiologia.umh.es\/?page_id=116"},"modified":"2025-09-21T19:54:35","modified_gmt":"2025-09-21T17:54:35","slug":"utilizacion-del-citrato","status":"publish","type":"page","link":"https:\/\/docenciamicrobiologia.umh.es\/en\/indice-de-practicas\/5-identificacion-bacteriana\/utilizacion-del-citrato\/","title":{"rendered":"Use of citrate"},"content":{"rendered":"<p><strong>Overview<\/strong><br \/>\nThis test enables us to distinguish between a group of bacteria that can grow with citrate as the only source of carbon and inorganic ammonium salts as the only source of nitrogen, which alkalizes the medium.<\/p>\n<p>The medium to use is Simmons&#8217; Citrate agar, which contains sodium citrate, ammonium phosphate and bromothymol blue as a pH indicator.<\/p>\n<p>Only the bacteria that are capable of metabolizing the citrate will be able to multiply in this medium and they will release ammonium ions which, together with the citrate metabolism, will produce a strong basification of the culture medium, which will become apparent with the change in color from green to blue.<\/p>\n<p style=\"text-align: center\"><iframe  id=\"_ytid_78409\"  width=\"480\" height=\"270\"  data-origwidth=\"480\" data-origheight=\"270\" src=\"https:\/\/www.youtube.com\/embed\/d2uiwBF2jbY?enablejsapi=1&autoplay=0&cc_load_policy=0&cc_lang_pref=&iv_load_policy=1&loop=0&modestbranding=0&rel=1&fs=1&playsinline=0&autohide=2&theme=dark&color=red&controls=1&\" class=\"__youtube_prefs__  no-lazyload\" title=\"YouTube player\"  allow=\"accelerometer; autoplay; clipboard-write; encrypted-media; gyroscope; picture-in-picture\" allowfullscreen data-no-lazy=\"1\" data-skipgform_ajax_framebjll=\"\"><\/iframe><\/p>\n<p><strong>Carrying out the practical<\/strong><br \/>\nThe slant agar culture is inoculated on the surface by means of an inoculation loop.<\/p>\n<p>Incubate at 37\u00baC for 24 hours.<\/p>\n<p>After the incubation period the results are read.
