Material

• Microscope slide with a heat fixed sample
• Set of dyes. Malachite green and Safranin.
• Swab to flame.
• Metal tweezers.
• Stopwatch.

Procedure

1. Cover the microscope slide with the malachite green.
2.Heat until vapor is released. Carry on for 3 minutes.
3.Wash with water.
4.Cover the microscope slide with safranin to compare. Leave it for 1 minute.
5.Wash with water and dry with filter paper.

In the microscopic observation the spores are stained green and the vegetative cells are pink.
